The test is based on the binding of the adenovirus antigen to immobilized antibodies. Binding is detected through peroxidase-labelled anti-adenovirus antibodies.
 
The ELISA for the detection of respiratory adenovirus recognizes conserved epitopes. Therefore, it may also find use outside of human medicine in some areas of veterinary medicine.

The test for adenovirus antigens in human stool samples has been optimized to detect the major serotypes 31, 40 and 41. The monoclonal antibodies of the test system detect conserved epitopes of the hexon protein.
